Formaldehyde-Agarose RNA gel


 
 

10X MOPS electrophoresis buffer.
0.2 M MOPS (pH: 7.0)
20 mM Sodium Acetate
10 mM EDTA (pH:8.0)
 

1. Dissolve 41.8 g MOPS in 700 ml DEPC-H2O.
2. Adjust pH to 7.0 with NaOH
     3. Add 20 ml of DEPC-treated 1M Sodium Acetate.
(1M Sodium Acetate:Dissolve 41g Sodium Acetate (FW:82.03)
in 400 ml DEPC-H2O.Adjust volume to 500ml.)
                         4. Add 100 ml of DEPC-treated 0.1M EDTA (pH:8.0)
                     ( 0.1 M EDTA: dissolve 18,61 g EDTA (FW:372.24) in 400 ml DEPC-H2O,
                                  use magnetic stirrer. Adjust pH to 8.0 with NaOH.Adjust to volume  500 ml.)
5.Adjust volume to 1000 ml with DEPC-H2O.
   6. Sterilize solution by filtration through a 0.45um
   Millipore filter.
 7.Store at roomtemperature.
 
 
 

Loading buffer 50 ml.
1. 1 mM EDTA, pH:8.0 (500ul 0,1M EDTA)
2. 0.25% (w/v) Bromphenol blue ~0.1g
3.0.25% (w/v) Xylene cyanol ~0.1g
4. 50% (v/v) Glycerol 25 ml
5. H2O  24.5 ml
