Tm analysis

Tm analysis is a method for extracting rough sequence information from qPCR analysis.

The dissociation curve step, performed after the amplification cycling in the qPCR machine is complete, gives the melting temperature (Tm) of your amplicons. Differences in Tm imply differences in amplicon sequence.

GcTm was developed to improve Tm determination in data aquired from and ABI Prism 7000. This in conjunction with a Tm-probe can improve resolution of Tms by three fold. Please refer to the publication "Molecular beacon based temperature control and automated analyses for improved resolution of melting temperature analysis using SYBR I Green chemistry – Improved melting temperature determination. Christoffer Nellåker, Ulf Wållgren and Håkan Karlsson. Clinical Chemistry" for more information. GcTm can be downloaded here.
